Yes you can.

My busiest system has 3000 users actively using the system, and we backup the databases without a problem.

We run daily full backups and transaction log backups every 15 minutes.

It can affect performance, just depends how your hardware is setup, specifically the disks.

You should look into a third-party backup utility that can compress it while it is backing up, saving disk space and backup time. We use Red Gate's SQL Backup (cheap) and Quest's Litespeed (not so cheap).

For our 2008 systems using Enterprise edition, we use native compression.

Compressing your backups saved 75-90% of disk space and around 75% of backup time. You should really look into this.

By the way on the system that I mentioned, the database size is 1TB. We are able to backup that guy daily without any issues. We are backing up to a SAN that is super fast and are using Litespeed.
