I am the developer for a SQL Server, the Administrator is out on vacation. On Sunday, our web apps that reference a particular database became dramatically slower. Also, a DTS Package which normally takes minutes now takes hours. No new development took place after Friday. As far as I know, no administrative changes were made after Thursday.

I am not very familiar with the administration end, but I did make sure that there was space on the disk for both the database and its log, and that both were set to grow as needed.

Can you suggest a simple checklist of general items that I could run through to determine the cause? Bear in mind that I have only a basic understanding of the administration side.

Here are some things to check:

System monitor (Performance monitor) - check to see where the problem is
SQL Profiler - figure out which queries are problematic
DBCC SHOWCONTIG - check to see if fragmentation exists

The first one is pretty easy. The other two are going to require a little knowledge of them. Have you used SQL Profiler before? For DBCC SHOWCONTIG, just check the scan density on all of the indexes. If it isn't very close to 100%, you'll then want to reindex.

Maybe sp_who2 might help as well, see if you have any open connections that should be closed, as well as see if there are any connections blocking other connections.

Just do this in Query analyzer:
 exec sp_who2
