You cannot copy the files (.mdf, .ldf) without stoping the server or detaching the databases. Use Database Maintanence Wizard to schedule and backup the database.

Instead of using the Database Maintenance Wizard, just create your own BACKUP DATABASE commands so that you have more flexibility. Then schedule these commands by creating a job.
